1.A positive line appears infrequently when testing a sample with high concentration of food allergen proteins.

In case of containing the subject protein in extremely high concentration, a positive line does not appear even the confirmation window is colored (prozone phenomenon). If you wish to assay such a sample, dilute the sample appropriately to re-assay.

* According to samples, the procedures of diluting manipulation are different.

【In case of food inspection】: after the Sample Extract is diluted with Extraction Solution appropriately, dilute it 10-fold with Diluting Solution for examination.

【In case of swab test】: after the Sample Extract is diluted with Extraction Solution appropriately, dilute it 10-fold with Diluting Solution for examination.

2.The confirmation window is not colored red at the end of the reaction time after adding the Test Solution to the test stick.

This is because there is not enough Test Solution to develop, so additional dilution is recommended for re-assay.

3.The sample, such as thickened polysaccharides, is hard to dissolve in Extraction Solution thoroughly.

In this case, reduce the sample amount to fully dissolve at the extraction procedure for examination. However, due to the change of dilution rate at the extraction procedure, the detection sensitivity converted to the value will be different in case of food sample.

e.g. in case of “extracting 0.5 g of a sample with 19 mL of Extraction Solution (39-fold extraction)” instead of the manipulation of “extracting 1 g of the sample with 19 mL of Extraction Solution (20-fold extraction),” the detection sensitivity on the test stick is 25 ng/mL, which does not change, but the detection sensitivity converted to the value in case of food sample changes from 5 μg/g to 9.75 μg/g.
